Studies on Organogenesis

چکیده

1. Isolated blood cells, when placed for incubation in a plasma substratum, are capable of constructing highly organized, tubular processes that project from the explanted cell mass into the surrounding medium. 2. The tubular structures have fibrillar walls that may be covered, eventually, by a membranous layer of leukocytes. Their lumina contain blood cells suspended in fluid. 3. The formation of the tubules is initiated by the red cells. The leukocytes, and more particularly the thrombocytes, are responsible for the construction of the walls. 4. The phenomenon occurs only in the presence of plasma, the coagulation of which has been slightly delayed. Once the surrounding medium has become firmly coagulated, no further change occurs either in the length of the tubules or in their diameter. 5. The development of the tubules is not suppressed by substances that enhance cellular activity unless they induce, at the same time, the immediate coagulation of the medium. The addition of embryo tissue juice prevents their formation by producing early coagulation. 6. The phenomenon as a whole is dependent upon the physicochemical nature of the medium, the character and thickness of the explanted fragment, and the physical and physiological peculiarities of the cells that comprise it. It is the expression of various physicochemical and physiological events that have occurred in a definite order, or sequence.

Somoclonal Variation Studies on Phyllanthus amarus Schum & Thonn

The present study was aimed at the development of a somoclonal variant and isozyme marker for Phyllanthus amarus Schum & Thonn using inter-nodal segments and the enzyme peroxidase. Maximum callus proliferation was obtained on Murashige and Skoog’s medium supplemented (MS) with 1.0 mg/l of 2,4-Dichlorophenoxyacetic acid. Three weeks-old pale yellowish white semi-friable callus was used for organ...

Efficient protocol for break impasses of regeneration via callus for 20 genotypes of chickpea

The results of studies on in vitro regeneration of chickpea (Cicer arietinum L.) have shown that indirect regeneration via callus was not successful. The introduction of an efficient and repeatable regeneration protocol is necessary for using transgenic technologies for the improvement of chickpea. The present study was undertaken to investigate the possibility of indirect regeneration in vario...

Callus Induction and Organogenesis Capacity from Lamina Explant of Petunia × hybrida F1 Induced by BA and NAA

Plant growth regulators (PGRs) have an important role in callus induction and organogenesis of plant explants cultured in vitro conditions. Callus has a proper potential for plant regeneration through indirect organogenesis and embryogenesis as well suspension culture, genetic transformation and production of secondary metabolites. In current experiment, leaf explants of Petunia × hybrida F1 we...

Roles of phospholipase C isozymes in organogenesis and embryonic development.

Phosphoinositide metabolism is an important intracellular signaling system that regulates a variety of cellular functions. Phospholipase C (PLC) is a key enzyme in this system. Recent studies on genetically manipulated mice have clarified the functions of PLC in vivo. This review focuses on the roles of PLC in organogenesis and embryonic development.

A Simple and Efficient Direct Shoot Organogenesis Method Using Leafy Petiole Explants in Gerbera jamesonii ‘Royal Soft Pink’

The gerbera market would benefit from an efficient and simple protocol for high rate regeneration for propagation and genetic engineering. With these objectives, this investigation was done on shoot regeneration via direct organogenesis from leafy petiole explants of Gerbera jamesonii ‘Royal Soft Pink’. Murashige and Skoog (1962) (MS) medium was supplemented with 0.1 mg L-1 indole-3-acetic acid...
